Initially set up in 1992 in the Department of Genetics of the Necker Hospital, the DNA-Biological Resources Center (BRC)’s aim is to perform long term and safety storage of precious biological samples under various forms (DNA, leukocyte pellets, Peripheral Blood Lymphocyte or EBV-transformed cell lines) from blood and fibroblasts culture of patients and their families affected with rare genetic disorders, in the current state of the art conservation and distribution procedures, in compliance with national and European regulations for biobanking.
Two types of samples are stored in the Necker DNA-BRC :
- samples from patients (almost only children) with rare genetic disorders who are attending the various specialized departments of the Children hospital;
- samples which are part of collections generated through a defined research project
The DNA-BRC team, in collaboration with the local clinical Research unit, helps users in the constitution and declaration of sample collections, is part of national projects “National Infrastructure in Biology and Health” (Biobanks, Rare Diseases Cohorts, Hospital-University Institute) and of the european project Biobanking Biomolecular Resources Research Infrastructure (BBMRI).

The DNA Biobank mainly comprises a biosafety level II laboratory for blood sample processing and cell culture, one automated DNA extractor Autopure LS, QIAGEN (DNA sample quality has been successfully tested for exome and CGH array with the saline method used), one storage room equipped with -80°C and -140°C mechanical freezers with adequate CO2 and nitrogen back-up as well as multiple safety procedures.
In the Imagine Building, the centralized facility will be mainly composed with three biosafety level II laboratory for blood sample processing and cell culture, one large storage room equipped with -40°C, -80°C and -140°C mechanical freezers with adequate CO2 and nitrogen back-up as well as multiple safety procedures, and nitrogen storage. The automated DNA extractor will be localised in this plateform. All the Biological collections will be kept securely, their management and the quality of their preservation improved, with respect of the regulations and ethical recommandations.

Then, a meeting is organized to define together the practical modes of the project.
For the acceptance of the blood sample to the CRB, the following documents are necessary :
- an of the blood sample, the diagnosis is imperative
- a major consent or a minor consent or a fibroblasts culture consent.
In absence of these documents, a non-conformity will be generated.
Reminder : for a sediment of the white globules or an extraction of DNA, take a blood sample on an EDTA tube on ambient temperature.
For putting in line or lymphocytes, take a blood sample on citrate or ACD tube on ambiant temperature.
To make a release of a sample :
- A by mail (email@example.com) of the medical doctor legal responsible of the blood sample is necessary.

You have been kind enough to respond to the survey of satisfaction of DNA-BRC during the month of December 2014 , we thank you . We received 18% of responses: 100 % of those who responded were pretty informed during the period of the move.
MAJOR INFORMATION +++
Starting from Monday, December 15, 2014, given the very large number of samples that arrive at DNA-BRC, blood samples will now be systematically converted into leukocyte pellets and will be kept frozen. DNA extractions will be performed at the demand of release (requested by email or staff note for CGH) from the frozen white blood cells. An email will be sent to the requester when the DNA is available. The period shall be three weeks when the delay has been phased out.
The DNA-BRC does not perform emergency DNA extractions. However it is possible to dispose of leukocyte pellets in order to to perform the extraction by another departments. You just have to make a request by email and come pick the samples with dry ice at the DNA-BRC.

- Tuesday from 9am to 1pm and from 2 pm to 5 pm and for CGH
- Wednesday from 9 am to 12:30 pm
- Friday from 9 am to 12:30 pm
It is important to respect these times, because the increasing activity of the DNA-BRC does not longer allows us of the flexibility of before. If the outputs relate to a large number of samples (> 10), it is necessary to make an appointment so that samples can be prepared in advance.
